Q: What if I want to use other items with Cotton Root Barks?
A: Well there are certain items you can use with Cotton Root Barks. Let's talk about those now.
Lion Meat and GMO Cows can be used with Cotton Root Barks. But make sure to always use the Cotton Root Bark first, otherwise you are wasting a very expensive item. After using the Cotton Root Bark, you can use either Lion Meat or GMO Cow but not both. Why? Well that is because Lion Meat and GMO Cows both use up the same slot when the breeding is rolled. The roll order will be posted down below.
A few other items that can be used with Cotton Root Barks are as follows: Lion Scrotum (only on a mutated lioness (except hybrids), Buffalo Scrotum, Grain of Paradise, Great Tit, and Red Cocks.

Q: Can I use a Piebald/Patches Stud for this project?
A: You could but you would be negating the effect of the GMO Cow, Lion Meat and Cotton Root Bark. Piebald and Patches roll before these items. Which means any of the cubs that get the Piebald or Patches mutation will not get rolled for the GMO Cow, Lion Meat or Cotton Root Bark.

Q: Can I use a Primal or Primal Variant Stud for this project?
A: Yes. Primal and its variants roll after the items. So any cub that the roll didn't land on a mutation from the CRB, Fertility, GMO Cow or Lion Meat can possibly be a Primal or Variant.

Mutation roll order explained. This can also be found in the FAQ on Lioden, here - https://www.lioden.com/faq.php?cat=10#157.
† Note: this is only specifically for lionesses with one of these mutations, as they are heritable. If the lioness is a Leopon, it will roll for her mutation's natural pass chance first, then move on to Patches/Piebald if she was bred with a Patches/Piebald stud, etc. The same goes for Overgrown Fur - it will roll for her mutation's natural pass chance first, then move on to Patches/Piebald if she was bred with a Patches/Piebald stud, etc. Finally, the same goes for Tigons. Patches/Piebald will only roll with their own natural pass chance if either the lioness or the stud has that mutation.
If the mother does not have a heritable mutation, the official mutation roll order will operate the same as listed above, just without the chance to inherit Leopon, Overgrown Fur, Tigon, Patches, or Piebald (assuming she was not bred to a Patches/Piebald stud).
To clarify, heritable mutations will only roll if one of the parents has the mutation, and it can pass down from that parent. If you do not breed a lion with a heritable mutation, they will not "roll" when your cubs are conceived and generated. If you do not use mutation-boosting items, they will not "roll" when your cubs are conceived and generated. The mutation roll order above is only assuming that a lioness has a heritable mutation, is bred to a Dorsal Fur, Patches, Piebald, or Primal stud, mutation items have been used on her, etc.
* Note: the Tigon boost was only applicable until November 30th, 2018. Though the Tigon boost rolls third-to-last, it will overwrite all other mutations if it triggers. It is recommended that you do not use the Tigon boost on a lioness with a heritable mutation, as any potential Leopon, Overgrown Fur, Patches, Piebald, or Tigon cubs that may have been produced will be overwritten by the Tigon boost's activation.
* Note: the Dorsal Fur guarantee is only applicable until May 31st, each year. Since it is done each year. Though the Dorsal Fur guarantee rolls last, it will overwrite all other mutations if it triggers, even a Mukombero if it was used on a lioness. It is recommended that you do not use the Dorsal Fur guarantee on a lioness with a heritable mutation, as any potential Leopon, Overgrown Fur, Patches, Piebald, or Tigon cubs that may have been produced will be overwritten by the Dorsal Fur guarantee's activation. You should also avoid using the Dorsal Fur guarantee on a lioness that has been given any mutation items (especially one as expensive as a Mukombero), as it will completely overwrite any mutated cubs and force them to have Dorsal Fur.
